Part II of Volume 1 focuses on approaches used to study megakaryocyte function, including the development of specialized structures for future production of platelets (e.g., the demarcation membrane system), the appearance of platelet-specific surface receptors, and the increase in ploidy. The source of megakaryocytes is often a complex issue facing many researchers owing to the extremely low density (<1%) of this cell type in its primary location, the marrow. Techniques to purify megakaryocytes from marrow based on their unique size and surface markers are described in Chapter 22, vol. 1, along with approaches to maintain these cells in culture and monitor formation of platelet-generating proplatelet structures. An alternative approach to generating megakaryocytes is to grow them in culture from precursor cells as detailed in Chapter 23, vol. 1. This requires the presence of thrombopoietin (Chapter 26, vol. 1) acting through its receptor, c-Mpl, and normally other cytokines. The availability of systems to generate megakaryocytes in vitro provides a promising avenue to generate genetically modified platelets. Although there is no doubt that continuous megakaryocyte cell lines are useful for some studies of signaling in these cells, they have their limitations and the pros and cons are discussed in Chapter 27, vol. 1. Many basic and advanced techniques for the general study of cell signaling have been applied in studies to characterize the mechanisms of regulation of platelet function. These include ligand binding assays, the study of protein and lipid kinases and phosphatases, the analysis of lipid rafts in the regulation of cell signaling, the measurement of intracellular calcium levels, electrophysiological techniques, nitric oxide signaling, the use of venom proteins, and the internalization of proteins into platelets through permeabilization. These techniques and more are presented in Part II of Volume 2. In many respects the megakaryocyte is a giant platelet. Differences do occur in the arrangement of cellular organelles and cytoskeleton in the two cells, however megakaryocytes respond to platelet agonists such as ADP with full downstream functional responses (discussed in Chapters 1 and 16, vol. 2). Therefore, despite differences in ultrastructure, the megakaryocyte has earned its place as a sufficient, if not comparable model of platelet signaling. Many of the signaling techniques are therefore beginning to be applied to the megakaryocyte which, because of its size, is proving to be an extremely interesting model for platelet signaling, particularly using single cell approaches such as imaging and electrophysiology (see Chapters 16 and 17, vol. 2). Part III of Volume 2 is dedicated to recent advances in molecular techniques and post-genomic techniques and how they may be applied to the study of platelets and megakaryocytes. This section includes descriptions of how retroviruses may be used to express genes in primary megakaryocytes, the use of GFP-fusion proteins to study signaling in live cells, two-dimensional electrophoresis for platelet proteomics, the production of platelet cDNA libraries and the use of gene array technology. Although the main aim of the book is to include practical approaches to the study of platelets and megakaryocytes, a series of perspectives chapters are included (Part I, vol. 2). These chapters review the current understanding of platelet and megakaryocyte biology in addition to their discussions of important new developments and ex-perimental strategies. Many of the methods chapters also include further discussion and background on specific techniques. This book has only been made possible by the efforts of many international experts in the field. We are grateful to them for their willingness to contribute their knowledge, in particular their tricks of the trade, which have resulted from many years of dedicated hands-on work. Platelets and Megakaryocytes_ Volume 1_ Functional Assays

Preface The average human body has in the order of 1012 circulating platelets. They are crucial for hemostasis, and yet excessive platelet activation is a major cause of morbidity and mortality in western societies. It is therefore not surprising that platelets have become one of the most extensively investigated biological cell types. We are, however, far from understanding precisely how platelets become activated under physiological and pathophysiological conditions. In addition, there are large gaps in our knowledge of platelet production from their giant precursor cell, the megakaryocyte. Understanding megakaryocyte biology will be crucial for the development of platelet gene targeting. The aim of Platelets and Megakaryocytes is therefore to bring together established and recently developed techniques to provide a comprehensive guide to the study of both the platelet and the megakaryocyte. It consists of five sections split between two volumes. The more functional assays appear in Volume 1, whereas Volume 2 includes signaling techniques, postgenomic methods, and a number of key perspectives chapters. Part I of Volume 1, Platelets and Megakaryocytes: Functional Assays, describes many well established approaches to the study of platelet function, including aggregometry, secretion, arachidonic acid metabolism, procoagulant responses, platelet adhesion under static or flow conditions, flow cytometry, and production of microparticles. Although one would ideally wish to perform experiments with human platelets, studies within the circulation using intravital microscopy require the use of animal models, which are described in Chapter 16, vol. 1. These approaches are becoming increasingly important in our understanding of how platelet responses contribute to the complex formation of thrombi within the circulation. Although naturally occurring genetic mutations can indicate the importance of specific proteins, these are limited in frequency and scope and thus many laboratories are using transgenic animals to delete or upregulate individual gene products (see Chapter 2, vol. 2). Consequently, the application of platelet techniques to murine models has become a focus of many labs in recent years (e.g., Chapters 2, 16, 20, vol. 1). In addition to basic and advanced approaches to study platelet function, several chapters in this section (particularly 1 and 2, vol. 1) focus on the long-standing issue of the effects of different anticoagulants and procedures to prepare platelets. The experimenter has a choice of studying platelets within the blood, in plasma, or in an artificial medium. In whole blood, potential interactions with other cell types and plasma proteins are included, which is in many ways the most physiological in vitro approach (see Chapter 6, vol. 1), however this is a complex situation and interpretation can be difficult. In studies within plasma, other cells are removed, but the clotting cascade is retained (see Chapter 5, vol. 1). Frequently, however, platelets are studied in isolation from other cells and plasma following their resuspension in an artificial medium. The preparation of platelets from human and other species is not a trivial matter and great care is required to ensure that the method of preparation does not adversely affect subsequent analysis (see Chapter 2, vol. 1).

I KNOW a married man with two children who bought a twenty-one-room motel near Denver many years ago in order to become its resident voyeur. With his wife nearby to assist, he cut rectangular-shaped holes in the ceilings of a dozen rooms, each hole measuring six by fourteen inches. Then he covered the openings with louvered aluminum screens that simulated ventilation grilles, but were, in fact, observation vents that allowed him, while he knelt or stood on the thickly carpeted floor of the attic, under the motel’s pitched roof, to see his guests in the rooms below. He continued to watch them for decades, while keeping an almost daily written record of what he saw and heard—and never once, during all those years, was he caught. I first became aware of this individual after receiving a hand-written, special delivery letter, without a signature, dated January 7, 1980, sent to my home in New York. It began: Dear Mr. Talese: Since learning of your long awaited study of coast-to-coast sex in America, which will be included in your soon to be published book, “Thy Neighbor’s Wife,” I feel I have important information that I could contribute to its contents or to contents of a future book. Let me be more specific. I am the owner of a small motel, 21 units, in the Denver Metropolitan area. I have owned this motel for the past 15 years, and because of its middle-class nature, it has had the opportunity to attract people from all walks of life and obtain as its guests, a generous cross-section of the American populace. The reason for purchasing this motel, was to satisfy my voyeuristic tendencies and compelling interest in all phases of how people conduct their lives, both socially and sexually, and to answer the age old question, “of how people conduct themselves sexually in the privacy of their own bedroom.” In order to accomplish this end, I purchased this motel and managed it personally, and developed a foolproof method to be able to observe and hear the interactions of different people’s lives, without their ever knowing that someone was watching. I did this purely out of my unlimited curiosity about people and not as just a deranged voyeur. This was done for the past 15 years, and I have logged an accurate record of the majority of the individuals that I watched, and compiled interesting statistics on each, i.e., what was done; what was said; their individual characteristics; age & body type; part of the country from where they came; and their sexual behavior. These individuals were from every walk of life. The businessman who takes his secretary to a motel during the noon hour, which is generally classified as “hot sheet” trade in the motel business. Married couples traveling from state to state, either on business or vacation. Couples who aren’t married, but live together. Wives who cheat on their husbands and visa versa. Lesbianism, of which I made a personal study because of the proximity of a U.S. Army Hospital to the motel and the nurses & military women who worked in the establishment. Homosexuality, of which I had little interest, but still watched to determine motivation and procedure. The Seventies, later part, brought another sexual deviation forward, namely “Group Sex,” which I took great interest in watching. Most people classify the foregoing as sexual deviations, but since they are practiced so commonly by the larger proportion of people, they should be reclassified as sexual interests. If sexual researchers & people in general could have the ability to see into other people’s private lives and see this practiced & performed, and to ascertain exactly how large a percentage of normal people indulge in these so-called deviations, their minds would change immediately. I have seen most human emotions in all its humor and tragedy carried to completion. Sexually, I have witnessed, observed and studied the best first hand, unrehearsed, nonlaboratory, sex between couples, and most other conceivable sex deviations during these past 15 years. My main objective in wanting to provide you with this confidential information, is the belief that it could be valuable to people in general and sex researchers in particular. Additionally, I have been wanting to tell this story, but I am not talented enough and I have fears of being discovered. It is hoped that this source of information could be helpful in adding an additional perspective to your other resources in the development of your book or future books. Off to Ashford to see the fair maid, heigh-ho, heigh-ho, Dunk thought miserably as he dug. When the hole was deep enough, he lifted the old man’s body in his arms and carried him there. He had been a small man, and slim; stripped of hauberk, helm, and sword belt, he seemed to weigh no more than a bag of leaves. Dunk was hugely tall for his age, a shambling, shaggy, big-boned boy of sixteen or seventeen years (no one was quite certain which) who stood closer to seven feet than to six, and had only just begun to fill out his frame. The old man had often praised his strength. He had always been generous in his praise. It was all he had to give. He laid him out in the bottom of the grave and stood over him for a time. The smell of rain was in the air again, and he knew he ought to fill the hole before it broke, but it was hard to throw dirt down on that tired old face. There ought to be a septon here, to say some prayers over him, but he only has me. The old man had taught Dunk all he knew of swords and shields and lances, but had never been much good at teaching him words. “I’d leave your sword, but it would rust in the ground,” he said at last, apologetic. “The gods will give you a new one, I guess. I wish you didn’t die, ser.” He paused, uncertain what else needed to be said. He didn’t know any prayers, not all the way through; the old man had never been much for praying. “You were a true knight, and you never beat me when I didn’t deserve it,” he finally managed, “except that one time in Maidenpool. It was the inn boy who ate the widow woman’s pie, not me, I told you. It don’t matter now. The gods keep you, ser.” He kicked dirt in the hole, then began to fill it methodically, never looking at the thing at the bottom. He had a long life, Dunk thought. He must have been closer to sixty than to fifty, and how many men can say that? At least he had lived to see another spring.
